Just wondering if anyone has suggestions for where I can buy secondhand drama CDs in Japan - not looking for online options, but in person shopping. Are there any well-known chain stores that deal with such items? Would bookstores carry them? I’m specifically looking for drama CDs of some of my favourite animes.
Book Off sells CDs and such as well as books, but the available range is entirely dependent on what people have donated, so there’s no guarantees.

Mandarake also sells CDs, so they should have drama CDs too, though selection would of course also be potluck

Mandarake (unlike Bookoff) have their physical-store stock catalogue online: e.g. this is the result of searching for ドラマ in their “CD” subcategory. The store name “SAHRA” is their online-shipping warehouse, but everything else is a real physical store (they have a map of where they are). Might be handy to check if there’s anything specific you’re after.
Google Maps may think Bookoff is a charity shop, but they do actually pay people for books

Aside from BookOff you can also try Suruga-ya which sells various secondhand anime goods (figures, merchandise, drama CDs and DVD/Blu-ray discs).
Do you mind sharing what sort of titles you are looking for? As others have said, BookOff may have some secondhand ones, but some locations may have higher potential of having the titles you are looking for. For general mainstream titles, BookOff has a big store near Shinjuku station. If you’re into titles more popular in the male otaku community, then try Akihabara locations. If you’re more into joseimuke anime popular with women then go to Ikebukuro. There’s also K-books voice actor in Ikebukuro which have a variety of (male) voice actor-related goods, which includes drama CDs.
Heads up that Surugaya in Kyoto is mostly just figures and games, I’m not sure if they have any CDs Kyoto Mandrake I’m pretty sure does have CDs (and like someone already said, does have their inventory online when in doubt).
